The genus of Crocus consists of 160 species worldwide, while in Greece the existence of 31 taxa, 12 of which are endemic, has been recorded. Crocus sativus is the only one cultivated for its styles, which constitute the well known saffron. Saffron contains a plethora of components like flavonoids and crocins, which could be of value to the treatment of cognitive impairment and other age related disorders. Although many studies have been conducted on C. sativus, little is known about the phytochemical composition of the other indigenous crocuses and their biological properties. In this study, we focused on Crocus nivalis from Panahaiko mountain (Achaia, Peloponnesus) which is an endemic species of the Balkan area and blooms in spring. In our lab, we studied the profile of volatiles extracted with diethylether by GC-MS, and the identity of polar metabolites extracted with methanol/water, 1/1 v/v, by HPLC. We analyzed in total three samples of C. sativus, one from the area of Kozani in Greece and two from the areas of Birgand and Quaenat respectively, that are located in Iran. None of the main known volatile components which were identified in C. sativus, was detected in C. nivalis. In addition, we founded that that the differences between the Greek samples and the Iranian ones of C. sativus are predominantly quantitative but not qualitative. The chromatographic analysis revealed not only quantitative differences in style concentration of known crocins, but also important qualitative differences due to the appearance of many peaks not present in C. sativus. In order to characterize the components, those were isolated by semi-preparative HPLC and the pure compounds were studied by Mass Spectrometry. As a result, 10 flavonol glycosides (mostly of kaempferol) and 10 crocins were identified. We show for the first time that the main crocin in C. nivalis styles is trans-crocin 6, which has not been previously reported in styles of other Crocus taxon. We also show the presence of a new crocin 3 with unique chemical structrure. The resuts of our lab could contribute to a wider understanding of the chemodiversity of the Crocus genus and potential therapeutic uses.el
