The aim of the present Master’s thesis was to investigate and optimize the formulation and processing parameters for the development of cod liver oil nanoemulsions enriched with the fat-soluble antioxidant Vitamin E, intended for topical dermal applications. Cod liver oil is characterized by a high content of omega-3 polyunsaturated fatty acids (PUFAs), particularly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), which are associated with anti-inflammatory activity and skin barrier restoration. However, its pronounced susceptibility to oxidative degradation necessitates its incorporation into advanced delivery systems capable of preserving its structural and functional integrity. The experimental work systematically evaluated the influence of formulation composition and processing conditions on the physicochemical stability, oxidative integrity, and biocompatibility of the developed nanoemulsions. High-Pressure Homogenization (HPH) was employed as the primary nanoemulsification technique and was comparatively assessed against ultrasonication. The findings demonstrated that HPH at 800 bar produced nanoemulsions with superior structural characteristics, achieving droplet sizes below 100 nm and an exceptionally low polydispersity index (PDI), indicative of high homogeneity and reproducibility. In terms of formulation design, the optimal surfactant ratio was identified as 2.5:1 (w/w) lecithin to PEG-15 hydroxystearate. This composition ensured high absolute zeta potential values (up to −82.60 mV) and provided combined electrostatic and steric stabilization, resulting in enhanced colloidal stability. The incorporation of Vitamin E significantly improved the antioxidant performance of the system, effectively protecting the sensitive PUFAs from peroxidative degradation without compromising the structural stability of the nanodispersions. Biocompatibility studies conducted on human dermal fibroblasts confirmed the favorable safety profile of the optimized nanoemulsions, with cell viability consistently exceeding 90% across the tested concentration range. Overall, this study demonstrates that the integration of rational formulation design and high-pressure homogenization enables the development of stable, functionally active, and biocompatible nanosystems. These nanoemulsions represent promising platforms for the formulation of innovative dermocosmetic and dermopharmaceutical products based on marine lipids.
