Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease that leads to progressive paralysis and death 5-10 years after the onset of symptoms. It is a multifactorial disease with currently undetermined etiology. Investigation of the genetic basis of the disease has revealed a variety of genes that contribute to its occurrence, with the most commonly affected genes being SOD1, FUS, TARDBP and C9orf72. This thesis is part of the program " Flagship Action for Neurodegenerative Disease Research at the Foundation of Precision Medicine" and the aim was to investigate genetic variants in regions of the SOD1, FUS and TARDBP genes in Greek patients with sporadic ALS (sALS). Sequencing 32 sALS patients yielded 6 variants in SOD1, 19 variants in FUS, and 37 variants in TARDBP. Several of these have been re-identified in previous studies, while many new ones were also found. Based on the in-silico evaluation of the variants’ pathogenicity, several characterized as pathogenic, while others as benign, because they have little influence on the gene. Of note is the identification of 2 new non-sense variants that lead to early termination of protein synthesis in the middle of the polypeptide chain of FUS (p.R241*) and TDP-43 (p.Y214*). The results of this study contribute to the global effort to elucidate the mechanisms and the genetic basis of the disease, leading to the development of a diagnostic panel that will contribute to the faster confirmation of the diagnosis of ALS or to the screening of individuals at high risk, such as the offsprings of patients.
