In recent years, an important part of bone research has been focused on the study of the close relationship between lipid metabolism and bone function. Studies have shown that mutations in the LDL receptor (LDLr) are associated with high plasma cholesterol levels and predispose to the occurrence of premature atherosclerosis in both humans and experimental animals. In the present study we investigated whether the LDL receptor insufficiency / absence affects the expression of molecules that play an important role in bone metabolism. Methodology: 12-week-old male mice with LDL- receptor deficiency (LDLr -/-) and wild type (WT) were used, all of them bred on a normal diet. In the two experimental groups of animals, whole bone marrow cells (WBMCs) were isolated by flushing from the femur and the bones of the tibia. Then we evaluate molecules involved in osteoblastic function (Runx2, Osx, RANKL, OPG), in osteoclastic function (TRAP, Cathepsin K) and in bone remodeling (RANKL, OPG, RANK, OPG / RANKL ratio), at mRNA level using qRT-PCR. Results: At the level of mRNA expression, the two major regulators of osteoblastic function, RUNX2 (p = 0.0135) and OSX (p = 0.007), were significantly elevated in LDLr-/- mice compared to WT. In contrast, osteoclastic markers and markers of bone remodeling such as Cathepsin K (p = 0.022), TRAP (p = 0.4278), RANK (p = 0.2318), RANKL (p = 0.1748), OPG 0.5843), and OPG / RANKL ratio (p = 0.072), were elevated in LDLr-/- mice, however, they did not show a significant statistical difference between the two experimental groups, with the exception of cathepsin K (p = 0.022) which prevalent in LDLr -/-. Conclusions: Lack of the LDL receptor has a positive effect on bone metabolism since it appears to increase the expression of molecules produced and secreted by osteoblasts. In addition, this deficiency induces osteoblastic function, and most likely also affects osteoclastic function indirectly, through osteoblastic effects on osteoclasts. However, osteoblastic versus osteoclastic function appears to predominate with an increase in bone mass in LDLr-/- versus WT mice. These findings improve the understanding of the correlation between bone metabolism and lipid. The proposed study is a timely and new approach to expanding the types of existing knowledge regarding the association of LDL metabolism with bone metabolism.el
