Atherosclerosis Related Disease (ARD) risk is clinically assessed by LDL cholesterol (LDL-C) levels compared to some, considered as normal, levels in the blood. LDL particle (LDL-P) concentration and size are only used in clinical research due to their complex and costly measurement. However, there has never been proved a direct causal relationship between ARDs and those indicators. Oxidized LDL (ox-LDL) is the only proven clinical cause-indicator for ARDs, but is also limited to clinical research because (i) its oxidative status is biochemically undefined, and (ii) it is quantified by non-specific/non-standardized assays. This study proposes the following objectives: 1. Isolate LDL by developing a fast, non-ultracentrifugal, method, and ox-LDL biochemical characterization with readily measurable, specific, oxidative markers, for a more reliable (clinical/research) assessment of ARD risk. 2. Complement the ox-LDL status indicators by the following indirect indicators in blood serum (in whole/fractionated blood, to be determined by the development of simple assays): (i) LDL-P concentration and size. (ii) LDL carotenoid content (as a new indicator for LDL antioxidant protection). (iii) Identification/measurement of oxidant and antioxidant factors in blood, which may contribute to ox-LDL levels, before and after statin treatment (in association with its known antioxidant properties). 3. Study the distribution of the new clinical indexes in healthy and susceptible to the emergence ARD people with type 2 diabetes, before and after statin treatment, and compare them with the distribution of LDL-C.el
