| el |
| dc.description.translatedabstract | Living bacteria contain a number of endogenous systems of toxin-antitoxin (TA) pairs that help them to survive in unfriendly environments. One of these systems is the MqsR-MqsA pair detected in the bacterium Escherichia coli. The mqsR gene encodes a ribonuclease toxin, whose protein expression inhibits the metabolism by inducing rapid termination of protein synthesis. The gene for the respective antitoxin (mqsA) encodes the antitoxin MqsA, which binds to MqsR and protects cells from its effects. Both genes constitute an operon which is repressed transcriptionally by the MqsA-MqsR complex. Under conditions of stress, MqsA is degraded while MqsR can exert its ribonuclease activity resulting in the formation of biofilms and persisters. The purpose of this work was the quest of additional functions of this TA pair. To this aim, the conditions for the overexpression and purification of MqsA, MqsR and MqsAN (the amino-terminal domain of antitoxin MqsA) were optimised, the proteins were immobilised on columns and cell exctracts were chromatographed on these columns. The eluted fractions were analysed by mass spectrometry. The overall picture suggests more functions for the particular TA pair. | el |
