Abstract

It is possible to design a method for phage display technology using whole cell panning in order to select binders with specificity for membrane proteins in their native conformation. In the case of CD3 selection, two unique clones that showed selectivity for CD3 protein were found and their production as free monobodies must be optimized in order to proceed with validation assays of their specificity and affinity. We also found a disadvantage about working with mutant libraries which consist of the risk of selecting clones that don’t correspond to full-length monobodies. Regarding the preliminary formulation studies of the LNPs that would be functionalized with the selected monobodies, it was noticed that the formulation is quite robust since the modification of parameters like temperature, buffer, and flow rate ratio didn’t produce big changes in the physical characteristics of most of the formulations. The optimal short-term storage condition is to keep the LNPs at 4 °C.
