Malaria is a serious infectious disease caused by parasitic protozoa of the genus Plasmodium, with high mortality rates, particularly in Southeast Asia and Africa. Although numerous antimalarial drugs have been developed, their indiscriminate and prolonged use has led to the emergence of resistant Plasmodium strains against classical therapies. In recent years, resistance phenomena have also been observed in artemisinin, which remains the most effective antimalarial drug. To address resistance, the World Health Organization recommends Artemisinin-based Combination Therapies (ACTs), where artemisinin is co-administered with other antimalarial agents, such as quinoline derivatives. Recently, G-factors (Growth Factors), which contain an endoperoxide bridge similar to that of artemisinin, have emerged as attractive targets for the development of synthetic analogues and the investigation of their promising antimalarial properties. As part of the present study, starting from syncarpic acid, the synthesis of three G3-endoperoxides (compounds 79, 81, and 83) was carried out, along with their further chemical modification through the introduction of appropriate functional groups, leading to the creation of six additional analogues (84–90), aiming at the evaluation of their antimalarial activity. Furthermore, these analogues bear suitable functional groups for the formation of hybrids (homo- or hetero-) via "click chemistry". In parallel, the artemisinin molecule was suitably modified at the C-10 position, yielding compounds 91, 92, 93, and 94, for subsequent conjugation with the G-endoperoxides. The biological evaluation of the final novel hybrid compounds is expected to provide important insights into the structure–activity relationship and contribute to the identification of new lead compounds against treatment-resistant Plasmodium falciparum parasites.
