In the attendance dissertation focuses on the study of the microbiome of the cultivated mussel in coastal areas of Greece using third generation sequencing techniques. The aim of the research is to examine how the health of the mussel is affected by the presence of pathogenic microorganisms, both for the organism itself and for the human consumer. In the first part of the study, a literature review on the biology and culture of Mytilus galloprovincialis is carried out. In particular, the general characteristics of the mussel, their breeding and cultivation methods are mentioned. Then, the mussel microbiome, its composition, its interaction with the organism and the effects of the environment are discussed. The Thermaikos Gulf, as an important mussel cultivation area, is analysed from both a geomorphological and environmental point of view, with emphasis on pollution. Subsequently, the evolution of DNA sequencing techniques is presented, with emphasis on modern methods of microbiome analysis, 16S rRNA sequencing and the use of the portable MinION device for real-time DNA and RNA analysis. The second part of the paper describes in detail the sampling steps, the isolation and the quantitative and qualitative evaluation of the DNA, as well as the bioinformatic analysis of the data. In addition, the electrophoresis and PCR techniques used to amplify and confirm the genetic data are discussed. The last part analyses the results revealing the composition of the mussel microbiome and the influence of environmental conditions on its formation. The diversity of microorganisms, their taxonomic distribution and the presence of pathogens that can affect both mussels and human health are presented. The main findings of the resulting investigation confirmed the existence of pathogenic microorganisms. The present work highlights the importance for further understanding of the mussel microbiome and the improvement of mussel culture in Greece.
