In the last decades, a large number of analytical methods have been developed to detect the adulteration of olive oil by other plant oils. The main categories include chromatographic and spectroscopic methods such as HPLC, GC, NMR, FT-IR and MS. However, the analytes detected by these methods are chemical components (metabolites), whose concentration and structure may vary between varieties, geographical areas of growth and environmental and weather conditions. This has led to an increasing interest in the development of molecular methods, as the DNA sequence of plant oil is exclusively linked to the species or varieties and is not influenced by environmental factors. Biosensors are one of the newest and most innovative strategies to detect adulteration of olive oil with other plant oils. In this Master's thesis, a new molecular method was developed that allows the detection of olive oil adulteration with other plant oils. The method involves three steps DNA isolation from five plant oils, PCR amplification using a plant-oil specific primer mixture and finally, detection of the PCR products with a multiple dry reagent strip type biosensor. The presence of adulteration is confirmed by the appearance of a red spot on the test zone where oligonucleotide probes identifying the specific adulterant species have been immobilised. Initially, the selectivity of the PCR reaction was investigated and the conditions for the plant-oil discrimination of both the PCR and the biosensor were optimised. Then, three types of olive oil adulteration detection (double, triple and multiple) were studied by developing the dry reagent strip type biosensor. The proposed DNA biosensor is specific, simple in design and use and provides fast results (15-20 min). It showed very good detectability, ability to detect adulteration rates as low as 5% for sesame, soybean and sunflower oils and up to 10% for corn oil. Finally, it showed excellent repeatability across all types of detection of olive oil adulteration with quite low %CV values.
