Dengue virus is a global health threat with an estimated 2.5 billion people in more than 100 tropical and subtropical countries at risk of infection. The virus is a member of the family Flaviviridae and genus Flavivirus and is related to the Zika, West Nile, yellow fever and Japanese encephalitis viruses. There are 4 serotypes of the virus: DENV-1, DENV-2, DENV-3, DENV-4 which are mainly transmitted through nygma by female Aedes aegypti mosquitoes. However, to date there are no approved drugs or vaccines against this virus. Current research efforts focus on inhibitors of NS2B-NS3 protease and NS5 polymerase, as their enzymatic activities have been shown to be essential in viral replication. Our research group, after structure-activity relationship (SAR) studies, discovered active amidoxime derivatives attached to a carbazole skeleton and more specifically, the di-amidoxime derivative SP-471P (EC50 1.10 μM, CC50> 100 μM) which acts as a pro - di-amidine drug SP-471 and is one of the most active and safest allosteric inhibitors in the literature against Dengue virus. Subsequently, the simplification of carbazoles to indoles was investigated, from which the conformationally more rigid indolazepinones. Derivative SP-1769B, demonstrated safety (CC50>100 μM) and one of the best antiviral activities against all DENV serotypes (DENV2/3 EC50 100 nM, DENV1/4 EC50 0.95-1.25 µM) in the literature. Further simplification of this molecule led to a pyrimidine series from which the derivative SP-393D molecule emerged as the best from this effort with EC50 = 0.11 µM, CC50 > 100 µM for DENV-2 and with improved clogP=3.19. The aim of this research work concerned firstly the synthesis of pyrimidine derivatives with improved activity and secondly with the exploration of the scaffold hopping from indolazepinone SP-1769B to purine derivatives that also led to novel potent Dengue antiviral compounds.
